Why does my reaction time vary between rounds?

Attention fluctuates, and so does the moment in your monitor refresh cycle when the colour changes. That is why we average five rounds instead of trusting a single attempt.

Does my monitor affect the result?

Yes. A 60 Hz display adds up to 16 ms of frame delay compared with a 144 Hz or 240 Hz panel, and input lag on the mouse and display adds a few more milliseconds.

Can reaction time be improved?

Somewhat. Sleep, caffeine, warming up and consistent practice all help, but the neural floor for visual reaction sits near 150 ms and no amount of training goes below it.
